[摘要] 目的 筛选人参皂苷及多糖最优化微波提取工艺。 方法 通过单因素实验和响应曲面法确定人参皂苷及多糖的微波辅助提取最佳工艺条件。 结果 人参皂苷的最佳提取条件为:固液比为1∶40,乙醇浓度为55%,微波提取时间为4 min,微波提取温度为60℃;人参多糖的最佳提取条件为:固液比为1∶50,微波提取时间为2 min,微波提取温度为90℃。按照优化后工艺条件实验,人参皂苷及多糖提取率分别为12.71%和23.32%。 结论 优化后的工艺稳定,微波辅助提取人参皂苷及多糖提取时间短,提取率高。

Optimization of microwave assisted extraction of Ginsenosides and Polysaccharide by respone surface methodology

[Abstract] Objective To screen optimal extraction process of ginsenosides and polysaccharides. Methods Single factor experiment and response surface method were applied and the optimum conditions were finally determined. Results The optimum extraction conditions of ginsenosides were: solid-liquid ratio was 1:40, ethanol concentration was 55%, microwave time was 4 min, microwave temperature was 60℃; the optimum extraction conditions of polysaccharides were: the solid-liquid ratio was 1∶50, extraction time was 2 min, the extraction temperature was 90℃. According to the optimized experimental conditions, ginsenosides and polysaccharide extraction rates were 12.71% and 23.32% respectively. Conclusion The optimized process is stable, microwave-assisted extraction of ginsenosides and polysaccharide are showed less time needed and the higher extraction rate.

人参为五加科多年生草本植物人参(Panax ginseng C. A.Mey.)的干燥根,为中华民族传统中药,有着4000多年的应用历史,是东北三宝之一。在悠久的中医药文化历史中,《神农本草经》首先记载了这一珍贵药材,堪称上品。据记载,人参具有“补五脏、安精神、定魂魄、止惊悸、除邪气、明目开心益智、久服轻身延年”的功效[1]。人参的主要有效成分人参皂苷具有清热解毒,促进肿瘤细胞的凋亡和分化[2],延缓衰老[3],调节中枢神经系统[4],提高机体免疫力[5],改善心脑血管供血不足等作用。人参多糖可明显增强免疫系统的功能并有抗肿瘤活性[6]。常用的中药有效成分提取法有浸渍法[7]、渗漉法[8]、煎煮法[9]、回流法[10]、索氏提取法[11]、超声波法[12]、超临界CO2萃取法[13]、超高压法[14]等。这些方法提取率一般较低,提取时间较长且能耗较大。微波辅助提取法是由微波提取和溶剂提取结合而成的一种新的提取技术,提取时间短、提取效率高并且重现性较好。

随着现代医药科学的发展研究,对人参主要活性成分人参皂苷及多糖的研究日益深入。在传统的人参有效成分提取中,人参皂苷和多糖的提取往往是单独进行的,本实验将提取人参皂苷后的人参渣经干燥后用于人参多糖的提取,实现了人参原材料的充分利用。本研究通过单因素实验考察了乙醇浓度、提取时间、提取温度、液固比对人参皂苷提取率的影响;研究了提取时间、提取温度、液固比对人参多糖提取率的影响;通过响应曲面中的Box-Behnken法优化了提取工艺,得到微波辅助提取人参皂苷及多糖的优化工艺参数。比较了水浴回流和微波辅助对人参皂苷和多糖的提取。
